    Ciliogenesis and the DNA damage response: A stressful relationship

    Both inherited and sporadic mutations can give rise to a plethora of human diseases. Through myriad diverse cellular processes, sporadic mutations can arise through a failure to accurately replicate the genetic code or by inaccurate separation of duplicated chromosomes into daughter cells. The human genome has therefore evolved to encode a large number of proteins that work together with regulators of the cell cycle to ensure that it remains error-free. This is collectively known as the DNA damage response (DDR), and genome stability mechanisms involve a complex network of signalling and processing factors that ensure redundancy and adaptability of these systems. The importance of genome stability mechanisms is best illustrated by the dramatic increased risk of cancer in individuals with underlying disruption to genome maintenance mechanisms. Cilia are microtubule-based sensory organelles present on most vertebrate cells, where they facilitate transduction of external signals into the cell. When not embedded within the specialised ciliary membrane, components of the primary cilium's basal body help form the microtubule organising centre that controls cellular trafficking and the mitotic segregation of chromosomes. Ciliopathies are a collection of diseases associated with functional disruption to cilia function through a variety of different mechanisms. Ciliopathy phenotypes can vary widely, and although some cellular overgrowth phenotypes are prevalent in a subset of ciliopathies, an increased risk of cancer is not noted as a clinical feature. However, recent studies have identified surprising genetic and functional links between cilia-associated proteins and genome maintenance factors. The purpose of this mini-review is to therefore highlight some of these discoveries and discuss their implications with regards to functional crosstalk between the DDR and ciliogenesis pathways, and how this may impact on the development of human disease

    To date, research involving functional neuroimaging of typical and atypical development has depended on several assumptions about the postnatal maturation of the brain. We consider evidence from multiple levels of analysis that brings into question these underlying assumptions and advance an alternative view. This alternative view, based on an “interactive specialization” approach to postnatal brain development, indicates that there is a need to: obtain data from early in development; focus more on differences in interregional interactions rather than searching for localized, discrete lesions; examine the temporal dynamics of neural processing; and move away from deficits to image tasks in which atypical participants perform as well as typically developing participant

    Dissolution influences on gypsum rock under short and long-term loading: Implications for dams

    Dissolution of soluble substrates such as gypsum presents a major hazard to dams in many parts of the world. This research simulates hypothesised conditions beneath the Mosul Dam, northwest Iraq, where collapse of a karstic system associated with continuous fresh water supply from its reservoir is a recognised problem. Gypsum rocks from northern Iraq and similar rocks from Bantycock gypsum mine, UK, were analysed for short-term mechanical response following immersion (5 to 50 weeks) and long-term loading during immersion (maximum 50 weeks). New experimental devices were developed from a conventional oedometer. Cylinder samples provided a proxy for massive gypsum strata. Samples were permanently submerged at atmospheric water pressure, with groundwater recharge, flow and dissolution simulated by regular changes of water. Stress on each sample was progressively increased to a maximum of 2688 kPa. Small increases in strain were recorded by the end of each test but no failures occurred within 60 days. However, notable failure due to atmospheric water pressure and axial stress occurred over long time periods. Visible physical changes included a decrease in sample mass and volume. Similar change was recorded in ultrasonic velocities. These indicate that gypsum collapse risk beneath dams requires prolonged exposure to dissolution. The modified device performed well and was robust, and demonstrates that such a modification can provide a simple low cost system for conducting laboratory creep tests on weak rocks

    Mathematics difficulties in extremely preterm children : evidence of a specific deficit in basic mathematics processing

    Background: Extremely preterm (EP, <26 wk gestation) children have been observed to have poor academic achievement in comparison to their term-born peers, especially in mathematics. This study investigated potential underlying causes of this difficulty. Methods: A total of 219 EP participants were compared with 153 term-born control children at 11 y of age. All children were assessed by a psychologist on a battery of standardized cognitive tests and a number estimation test assessing children’s numerical representations. Results: EP children underperformed in all tests in comparison with the term controls (the majority of Ps < 0.001). Different underlying relationships between performance on the number estimation test and mathematical achievement were found in EP as compared with control children. That is, even after controlling for cognitive ability, a relationship between number representations and mathematical performance persisted for EP children only (EP: r = 0.346, n = 186, P < 0.001; control: r = 0.095, n = 146, P = 0.256). Conclusion: Interventions for EP children may target improving children’s numerical representations in order to subsequently remediate their mathematical skills

    Lifting the veil of depression and alcoholism in sport coaching: how do we care for carers?

    The purpose of this article is to explore the insights of an elite sport coach living with comorbid depression and alcohol misuse. Such consideration is necessary because as coaching is increasingly repositioned as a caring activity the wellbeing of coaches themselves has rarely been considered. To address this gap, a narrative analysis methodology and a story telling approach was used to present the experiences of a case study coach (Steve). The story is derived from Steve’s own perspective and the perspective of his wife, Jane. This novel multi-voiced approach reveals the complex interplay between the sporting environment, Steve’s depression and his alcohol related problems. In doing so, Steve’s story makes an original contribution by unveiling the issue of mental health in elite sport coaching. Steve’s story depicts the significant impact of depressive symptoms, coupled by excessive alcohol use on a coach, their personal life, and their career. Using the work of Goffman and Sartre, the story alludes to how such symptoms can be hidden, to greater and lesser extents, from professional colleagues. This is an important theoretical contribution, because to care for coaches, colleagues, employers, and health professionals will need to understand the needs of individuals such as Steve. Developing an open and supportive culture, which accepts that coaches are fallible, may however be a challenge within professional sport contexts

    Reflected Light from Sand Grains in the Terrestrial Zone of a Protoplanetary Disk

    We show that grains have grown to ~mm size (sand sized) or larger in the terrestrial zone (within ~3 AU) of the protoplanetary disk surrounding the 3 Myr old binary star KH 15D. We also argue that the reflected light in the system reaches us by back scattering off the far side of the same ring whose near side causes the obscuration.Comment: 22 pages, 5 figures.
